Melanotan II (MT-II) is a synthetic analogue of the naturally occurring alpha-melanocyte stimulating hormone (α-MSH). It binds to melanocortin receptors throughout the body, stimulating melanin production in the skin, which can increase pigmentation and enhance tanning when combined with ultraviolet (UV) exposure. Researchers have also investigated its effects on appetite regulation and sexual function due to its activity on multiple melanocortin receptor subtypes.

Clinical and preclinical studies have demonstrated that MT-II can increase skin pigmentation by stimulating melanocytes to produce melanin. This may reduce the amount of UV exposure required to achieve a tan, although it does not eliminate the risks associated with UV radiation or provide complete protection against sun damage or skin cancer. Reported side effects in research settings include nausea, facial flushing, fatigue, increased libido, and darkening of existing freckles or moles. Long-term safety has not been established.

Melanotan II is currently an investigational peptide and has not been approved by Health Canada or the U.S. FDA for tanning, weight management, or any other general medical use. Additional human research is required to better understand its long-term safety, efficacy, and potential therapeutic applications.
